About Water DNA Extraction Kit

  • SPINeasy DNA Kit for Water is a high-performance water genomic DNA (gDNA) extraction kit based on silica-membrane spin-column technology. This kit enables quick isolation of gDNA from water in less than 30 min. Water samples are processed using our uniquely formulated Lysis Buffer W1 and Lysing Matrix E to effectively lyse various types of cells. Column W1 provided in the kit has high binding capacity and selectivity for gDNA. The combination of components in the kit extracts gDNA of high yield and purity that is ready for downstream analyses such as PCR, restriction digestion and sequencing.

Advantages of SPINeasy DNA Kit for Water

    Highly pure genomic DNA extraction

SPINeasy DNA Kit for Water is compatible with various types of water sample and resulting gDNA is of high purity. SPINeasy DNA Kit for Water provided better purity of extracted DNA when compared to the other DNA kits. The purity of the extracted DNA was determined with spectrophotometric analysis by measuring and calculating the ratio of the absorbance at 260 nm and 280 nm (A260/A280). Generally, purity is considered good with A260/A280 ratio around 1.7-2.0.

Note: The genomic DNA extracted with SPINeasy DNA kit for Water (in blue bar) was the only DNA kit among the competitors DNA kits to achieve A260/A280 ratio within 1.7-2.0 across different types of water sample.

The level of contaminants in the DNA extracted was determined similarly with spectrophotometric analysis by measuring and calculating the ratio of absorbance at 260 nm and 230 nm. Strong absorbance around 230nm can indicate that organic compounds or chaotropic salts are present in the purified DNA. A ratio of 260nm to 230nm can help evaluate the level of salt carryover in the purified DNA. The lower the ratio, the greater the amount of thiocyanate salt is present, for example. The contaminants may come the extraction procedure such as guanidine thiocyanate (GTC). Generally, purity is considered good with A260/A230 ratio of 1.0 or above 1.0.

Note: The genomic DNA extracted with SPINeasy DNA kit for Water (in blue bar) was generally free from the contaminants as assessed by its A260/A280 which were above 1.0 across different types of water samples.

Higher yield and integrity of genomic DNA extracted

SPINeasy DNA Kit for Water has consistently achieved better yield along with high purity as compared to competitors kits. Total yield is calculated by multiplying the DNA concentration by the final total purified sample volume and integrity is determined by absence of visible shearing or smearing of band during gel electrophoresis.

Note: SPINeasy DNA Kit for Water consistently achieved better yield across various types of water samples as compared to competitors DNA kits.

FAQ's of Water DNA Extraction Kit:

Q: How does the silica-membrane spin column method work in this Water DNA Extraction Kit?

A: The kit uses a silica-membrane spin column that binds DNA when water samples pass through under chaotropic salt conditions. Impurities are washed away while DNA is retained, then eluted with buffer to provide PCR-ready DNA suitable for analytical applications.

Q: What type of water samples can I use with this extraction kit?

A: This kit is compatible with a variety of water samples, including environmental, surface, ground, or drinking water. As long as the total volume does not exceed 1 liter, the protocol can efficiently extract DNA from most sample types.

Q: When should I use this DNA extraction kit in my laboratory workflow?

A: Use this kit whenever you need rapid, high-yield DNA extraction from water samples within one hour. It is especially valuable for time-sensitive PCR or molecular biology assays that require high-purity input DNA.

Q: Where should I store the Water DNA Extraction Kit for optimal performance?

A: Store the kit in a dry environment at room temperature, between 15-30C. No refrigeration is required, ensuring convenience and stability in most Indian laboratory or field settings.

Q: What is the process for extracting DNA using this kit?

A: Begin by filtering up to 1 liter of your water sample. Follow the protocol to lyse any captured cells, bind the DNA to the silica membrane, wash away contaminants, and elute pure DNA in a 50-100 L volume. The entire procedure typically takes less than 60 minutes.

Q: How do I benefit from using this Water DNA Extraction Kit?

A: The kit enables high DNA recovery rates, minimal processing time, and PCR-ready eluate. Its compatibility with large sample volumes and no requirement for cold storage makes it highly practical for both field and laboratory use, streamlining molecular research.
